Research Methods & Expected Outcomes

An efficient and reproducible regeneration protocol in castor was developed using hypocotyl explants of axenically grown seedlings. A successful transformation protocol in castor was also established along with the development of a Cas9 vector for transformation. In addition, a regeneration protocol in sunflower was standardized. All major laboratory and field infrastructure were strengthened, with the procurement of essential equipment such as -20°C and -80°C freezers, refrigerated mini centrifuge, refrigerated centrifuge, PCR machines (3 units), generator backup (45 KVA), fluorescence microscope, plant growth chambers (2 units), incubator shaker, laminar air flow units (2), and water bath shaker, as per the approved EFC. Furthermore, significant infrastructural improvements were carried out including repair and refurbishment of the tissue culture facility, molecular biology laboratory, transgenic greenhouse facility, and cold room. A new tissue culture room is being established, while chemicals and consumables required for the experiments have also been successfully procured.
