Research Methods & Expected Outcomes

We have designed a total of twelve guide RNAs for the selected targets (two for each, including homologues). Out of these, six have been cloned into the pBlu entry vector and confirmed through PCR and sequencing. From the pBlu entry vector, the guide RNA cassette was further cloned into the MDC123 binary vector, which was also PCR confirmed.

For standardization of the plant transformation protocol, we are using the Cas9 MDC123 empty vector along with Agrobacterium (EHA105)-mediated transformation. Regeneration protocols in all selected genotypes have been tested and optimized.
